They should go into effect immediately. However, if I'm not mistaken you will only get one notification per thread until the next time you visit.
By that, I mean if you were to subscribe to a thread that has had 18 responses since your last visit to that thread, you will get 1 notice, not 18. Once you visit that thread again, when someone else posts in the thread, you get another email notification.
I can't say for sure as I never use that option. I have way too many threads that I subscribe to that it would destroy my inbox.
